Evidence for a role of nitric oxide in hindlimb vasodilation induced by hypothalamic stimulation in anesthetized rats

Electrical stimulation of the hypothalamus produces cardiovascular adjustments consisting of hypertension, tachycardia, visceral vasoconstriction and hindlimb vasodilation. Previous studies have demonstrated that hindlimb vasodilation is due a reduction of sympathetic vasoconstrictor tone and to activation of beta2-adrenergic receptors by catecholamine release. However, the existence of a yet unidentified vasodilator mechanism has also been proposed. Recent studies have suggested that nitric oxide (NO) may be involved. The aim of the present study was to investigate the role of NO in the hindquarter vasodilation in response to hypothalamic stimulation. In pentobarbital-anesthetized rats hypothalamic stimulation (100 Hz, 150µA, 6 s) produced hypertension, tachycardia, hindquarter vasodilation and mesenteric vasoconstriction. Alpha-adrenoceptor blockade with phentolamine (1.5 mg/kg, iv) plus bilateral adrenalectomy did not modify hypertension, tachycardia or mesenteric vasoconstriction induced by hypothalamic stimulation. Hindquarter vasodilation was strongly reduced but not abolished. The remaining vasodilation was completely abolished after iv injection of the NOS inhibitor L-NAME (20 mg/kg, iv). To properly evaluate the role of the mechanism of NO in hindquarter vasodilation, in a second group of animals L-NAME was administered before alpha-adrenoceptor blockade plus adrenalectomy. L-NAME treatment strongly reduced hindquarter vasodilation in magnitude and duration. These results suggest that NO is involved in the hindquarter vasodilation produced by hypothalamic stimulation.

Helicobacter pylori chronic infection and mucosal inflammation switches the human gastric glycosylation pathways

Helicobacter pylori exploits host glycoconjugates to colonize the gastric niche. Infection can persist for decades promoting chronic inflammation, and in a subset of individuals lesions can silently progress to cancer. This study shows that H. pylori chronic infection and gastric tissue inflammation result in a remodeling of the gastric glycophenotype with increased expression of sialyl-Lewis a/x antigens due to transcriptional up-regulation of the B3GNT5, B3GALT5, and FUT3 genes. We observed that H. pylori infected individuals present a marked gastric local pro-inflammatory signature with significantly higher TNF-a levels and demonstrated that TNF-induced activation of the NF-kappaB pathway results in B3GNT5 transcriptional up-regulation. Furthermore, we show that this gastric glycosylation shift, characterized by increased sialylation patterns, favors SabA-mediated H. pylori attachment to human inflamed gastric mucosa. This study provides novel clinically relevant insights into the regulatory mechanisms underlying H. pylori modulation of host glycosylation machinery, and phenotypic alterations crucial for life-long infection. Moreover, the biosynthetic pathways here identified as responsible for gastric mucosa increased sialylation, in response to H. pylori infection, can be exploited as drug targets for hindering bacteria adhesion and counteract the infection chronicity.IPATIMUP integrates the i3S Research Unit, which is partially supported by FCT, the Portuguese Foundation for Science and Technology (PEst C/SAU/LA0003/2013). Estimating future streamflows is a key step in producing electricity for countries with hydroelectric plants. Accurate predictions are particularly important due to environmental and economic impact they lead. In order to analyze the forecasting capability of models regarding monthly seasonal streamflow series, we realized an extensive investigation considering: six versions of unorganized machines—extreme learning machines (ELM) with and without regularization coefficient (RC), and echo state network (ESN) using the reservoirs from Jaeger’s and Ozturk et al., with and without RC. Additionally, we addressed the ELM as the combiner of a neural-based ensemble, an investigation not yet accomplished in such context. A comparative analysis was performed utilizing two linear approaches (autoregressive model (AR) and autoregressive and moving average model (ARMA)), four artificial neural networks (multilayer perceptron, radial basis function, Elman network, and Jordan network), and four ensembles. The tests were conducted at five hydroelectric plants, using horizons of 1, 3, 6, and 12 steps ahead. The results indicated that the unorganized machines and the ELM ensembles performed better than the linear models in all simulations. Moreover, the errors showed that the unorganized machines and the ELM-based ensembles reached the best general performances

ABSTRACT Objective: This study aimed to report the experience of medication-related osteonecrosis of the jaws (MRONJ) in osteoporotic patients for nine years, and their associated initiating factors. Materials and methods: The numbers of invasive oral procedures (IOP) (tooth extraction, dental implant placement, and periodontal procedures) and removable prostheses performed from January 2012 to January 2021 were obtained from the digital records of a large public dental center. There were an estimated 6,742 procedures performed in patients under osteoporosis treatment. Results: Two cases (0.03%) of MRONJ were registered in nine years amongst patients with osteoporosis who had dental treatment at the center. From the 1,568 tooth extractions, one patient (0.06%) developed MRONJ. There was also one case from the 2,139 removable prostheses delivered (0.05%). Conclusions: The prevalence of MRONJ associated with osteoporosis treatment was very low. The protocols adopted seem to be adequate for the prevention of this complication. The findings of this study reinforce the rare frequency of MRONJ associated with dental procedures in patients submitted to the pharmacological management of osteoporosis. An integral analysis of systemic risk factors and oral preventive strategies may be considered regularly in the dental treatment of these patients

Post-harvest shelf life of fruits and vegetables is direct related to storage temperatures of these products. At controlled conditions of temperature and humidity, the speed of metabolic reactions can be reduced, allowing better conservation of the products. The evaluation was carried out in the Terminal Market of São Paulo- CEAGESP, to identify the condition of cold chambers used for storage of fruits and vegetables. The parameters investigated were: air temperature, relative humidity, refrigerated conditions, type of floor, chamber and door dimensions. Data were used to calculate thermal load. Apples and pears were the most stored commodities (63.25%) in CEAGESP, followed by bananas (24.10%). Based on the average commercialization volume, it was observed that 73.91% of the motors were super dimensioned.O tempo de vida pós-colheita de frutas e hortaliças está diretamente relacionado à temperatura de armazenamento do produto.
